This kind of thing could be translated easily into 5e as well, using a 5e background. Certainly in 4e it would be relatively trivial to come up with a set of 'Zodiac' themes, the player could simply pick one at character generation if desired to have his sign be a significant influence on his character's overall abilities. These provide a set of fairly small but significant static benefits, plus a single extra starting power, and a series of power swaps that allow you to pick some thematic powers if you want them (these are almost always 'utility' powers, which give you useful added capabilities or something similar). If you wanted a stronger kind of impact you could devise something akin to 4e's Themes. The benefit is small and limited in most cases, something like adding a skill to your list of trainable skills, getting a +2 bonus to a skill, knowing some facts about a specific subject that gets you a +2 if that becomes relevant in play, etc. This falls under 4e's Background mechanic. You could follow a rather more '4e-esque' approach and simply allow players to define a minor benefit they get for being 'born under a sign of X' where the player simply defines what that is (or the DM can tell them what the signs are in that setting and they can come up with some appropriate benefit).
